| Description | Nervegrowthfactorreceptor(NGFR)isalsoreferredtoasp75(NTR)duetoitsmolecularmassanditsABIlitytobindatlowaffinitynotonlyNGF(see162030),butalsootherneurotrophins,includingbrain-derivedneurotrophicfactor(BDNF;113505),neurotrophin-3(NTF3;162660),andneurotrophin-4/5(NTF5;162662).Atthetimeofitsdiscovery,NGFRwasconsideredauniquetypeofprotein.Subsequently,however,alargesuperfamilyoftumornecrosisfactorreceptorswerefoundtosharetheoverallstructureofNGFR(4extracellularligand-binding,cysteine-richrepeats,orCRs,andsignalingthroughassociationwith,ordisassociationfrom,cytoplasmicinteractors).TheidentificationofthissuperfamilyhelpedelucidatesomeoftheBIOLOGicfunctionsofNGFR,includingitsultimateinvolvementinthenuclearfactorkappa-B(NFKB;see164011)andapoptosispathways.Asamonomer,NGFRbindsNGFwithlowaffinity.Higheraffinitybindingisachievedbyassociationwithhighermolecularmass,low-affinityneurotrophinreceptors,namelythetropomyosinreceptorkinases,TRKA(NTRK1;191315),TRKB(NTRK2;600456),andTRKC(NTRK3;191316).TRKA,TRKB,andTRKCarespecificforor"preferredby"NGF,NTF5andBDNF,andNTF3,respectively.NTF3alsobindstoTRKAandTRKB,butwithsignificantlyloweraffinity. |
