Background: Phospholamban(PLB/PLN)isasmalltransmembraneproteinwhichplaysanimportantroleincontrollingtheactivityofthesarcoplasmicreticulumATPase(SERCA2a)ofcardiacmuscleduringcalciumsequestration.PhospholambanisphosphorylatedonseparateaminoacidresiduesbycAMP-dependent,andcGMP-dependent(Ser-16) andCa2+/CaM-dependent(Thr-17) proteinkinasesinresponsetoβ-adrenergicstimulation. AkthasalsobeenshowntophosphorylateThr-17.Theresultisanincreasedcalciumpumpactivitywhichreducesthetimecourseofthecalciumtransient,increasesthecalciumloadinthesarcoplasmicreticulum,andconsequently,producesalargercalciumtransientatthenextactionpotential.
