Theamine-modifieduridine5"-triphosphates(suchasaminoallyl-UTP)canbeusedtoproduceamine-containingRNAbyconventionalenzymaticincorporationmethodssuchastranscription,nicktranslation,randomprimedlabeling,orPCR.AminoallylUTPcanbereADIlyincorporatedintoRNAthroughtheconventionalenzymaticincorporationtechniques.Theresultingamine-modifiednucleicacidscanthenbelabeledusinganyofamine-reactivefluorescentdyes,biotinsandotheramine-reactivereagents.Theaminoallyl-modifiednucleotidescanbeincorporatedtoextremelyhighandconsistentlevelscomparedtothetag-labeleduridinetriphosphatesthatgenerallyhavehigherstereo-hindrance.Subsequentreactionoftheamine-modifiednucleicacidwithanexcessofamine-reactivereagentachievescorrespondinglyhighandconsistentlabelingefficiencies,regardlessofthelabelingreagentchosen.Thistwo-steplabelingmethodalsoeliminatestheneedtooptimizeanenzymaticreactiontoaccommodatedifferentdye-modifiednucleotides,whichmayincorporateatverydifferentrates.ThislabelingmethodiswidelyusedforbothFISHprobesandmicroarray-basedexperiments.
