Whenexposedtoexcitationlight,fluorescenceintensityofdyesdecreasesduetotheirphotooxidationorotherphotoreactions.Thereareveryfewfluorescentdyesthatcompletelyresistphotobleaching.Frequently,whenasectionhasbeenscannedrepeatedlyunderstrongexcitationlight,dyescouldlosesignificantfluorescencesignalbeforevisualevaluationorphotographycanbeaccomplished.Forexamples,thephotobleachingoffluoresceins(suchasFITC-labeledantibodies)hasbecomeamajorprobleminfluorescencemicroscopy.Inseverecases(suchasphycoprotein-labeledbioconjugates),afluorescenceimageofhighresolutioncannotevenbetakenduetotheextremelyhighphotobleachingrate.Fluoroquest™Anti-FADIngKitistoreducethedyephotobleachingrate,givingresearcherslongerobservationtime.Thekitcontainsalltheessentialcomponentsthatcanbereadilyappliedtoimagingexperiments.Theyareallpremixedandready-to-usesolutions.Thiskitisdesignedformicroplateformatwhile#20001isdesignedforslideformat.

Thisprotocolonlyprovidesaguideline,andshouldbemodifiedaccordingtoyourspecificneeds.

1.      MakeAnti-fadingsolution:AddtheentirevialofAssaybuffer(ComponentB)tothevialofAnti-fadingReagent(ComponentA).

Note:Aliquotandstoretheunusedanti-fadingsolutionat-20oC,protectedfromlight.Avoidrepeatedfreeze/thawcycles.ItisstillOKtousewhenthesolutiongoesbrown.Discardwhenthesolutionisblack.

 

2.      Applyanti-fadingreagent:Removeanyexcessliquidfromyour96-wellplate.Add100μLofAnti-fadingsolution(fromStep1)toeachselectedwells.

 

3.      Preparesamplesforimaging:SamplescanbeimagedimmediatelyafterapplytheAnti-fadingsolution.AtypicalimageisshowninFigure1.Storetheplateat4ºCinthedarkforoptimumsamplelongevity.
