NADPHprovidesthereducingequivalentsforbiosyntheticreactionsandforoxidation-reductioninvolvedinprotectionagainstthetoxicityofROS(reactiveoxygenspecies).NADPHisalsousedforanabolicpathways,suchaslipidsynthesis,cholesterolsynthesisandfattyacidchainelongation.Itisthesourceofreducingequivalentsforcytochrome450hydroxylationofaromaticcompounds,steroids,alcohols,anddrugs.NADPHisanecessarycofactorinmanyxenobioticmetabolismreactions.Inchloroplasts,NADPisreducedbyferredoxin-NADPreductaseinlaststepoftheelectronchainofthelightreactionsofphotosynthesis.TheNADPHproducedisthenusedasreducingpowerforthebiosyntheticreactionsintheCalvincycleofphotosynthesis.ManyoxidoreductasesandallligasesuseNADPHascoenzymes.NADPHisrequiredforthemeasurementofoxidaseactivitycatalyzedbyP450s,FMOs,NADPH-P450reductase,andmanyotheroxidaseenzymes.AATBioquest"sRediUse™NADPHRegeneratingKitprovidestwoready-to-usesolutionstoregenerateNADPHbyasimplemixing.ThiskitcanbeusedforallNADPH-requiringoxidaseassays(CDNA-expressedenzymesandliverfractions).About300-500enzymeassayscanbeperformedusingthiskit.Thetotalnumberofassaysthatcanbeperformeddependsonaresearcher"sexperimentaldesign.

  1. ThawComponentA,ComponentB,andComponentCatroomtemperaturebeforeuse.
  2. Make2XNADPHRegeneratingSolutionbyaddingthewholecontentofAssayBufferII(ComponentB)and500XGlucose-6-phosphatedehydrogenase(ComponentC)intoAssayBufferI(ComponentA),andmixthemwell.
  3. Addequalvolumeof2XNADPHRegeneratingSolution(fromStep2)intothedesiredassaysystem.
