D-Luciferingalactosidecontainsabeta-galactosidemoietyattachedatthe6-O-position,thuspreventingitberecognizedbythefireflyluciferaseenzyme.However,thegalactosemoietyisremovedeffectivelybybeta-galactosidaseactivity,andtheresultedD-luciferiniswellrecognizedbyluciferase.D-Luciferingalactosideisasensitivesubstrateforthechemiluminescentmeasurementofgalactosidaseactivityinhomogeneousassays,orincelllysatesamplesincombinationwithluciferaseanditscofactors.Whenusedasadualsubstrate,beta-galactosidase(LacZgene)andluciferaselevelscanbedeterminedwithhighsensitivity.Whenusedinamodelsystemexpressingfireflyluciferase,theluciferinisthenutilizedinafireflyluciferasereactiontogeneratelight.Itcanalsobeusedforinvivobioluminescenceimagingapplications.
