TheFBXL10ChemiluminescentAssayKitisdesignedtomeasureFBXL10activityforscreeningandprofilingapplications.FBXL10,alsoknownasKDM2B,JHDM1B,andCXXC2,isahistonelysinedemethylasethatexhibitsdemethylationactivitytowardtrimethyllysine4(H3K4me3)anddimethyl-lysine36(H3K36me2)onhistoneH3.ThekeytotheFBXL10ChemiluminescentAssayKitisahighlyspecificantibodythatrecognizesdemethylatedsubstrate.Withthiskit,onlythreesimplestepsonamicrotiterplatearerequiredfordetectionofdemethylaseactivity.First,FBXL10enzymeisincubatedwiththemethylatedH3peptideforonehour.Next,primaryantibodyisadded.Finally,theplateistreatedwithanHRP-labeledsecondaryantibodyfollowedbytheadditionoftheHRPsubstratetoproducechemiluminescencethatcanbemeasuredusingachemiluminescencereader.
