TheATAD2AInhibitorScreeningAssayKitisdesignedtomeasuretheinhibitionofATAD2Abindingtoitssubstrate.ThekeytotheATAD2AInhibitorScreeningAssayKitisaspecificbindingoftheATAD2Abromodomaintotheacetylatedhistonesubstrate.Withthiskit,onlythreesimplestepsonamicrotiterplatearerequired.First,asamplecontainingATAD2Aandaninhibitorofchoiceisincubatedwiththebiotinylatedsubstrateforthirtyminutes.Next,acceptorbeadsareadded,thendonorbeads,followedbyreADIngtheAlpha-counts.

Contraindications:
DMSOabove0.5%.OnlylimitedamountsofDMSOcanbeincluded,asithasbeenshowntodisruptBRD-ligandinteraction.AvoidgreenandbluedyesthatabsorblightintheAlphascreen®signalemissionrange(520-620nm),suchasTrypanBlue.Avoidtheuseofthepotentsingletoxygenquencherssuchassodiumazide(NaN3)ormetalions(Fe2+,Fe3+,Cu2+,Zn2+andNi2+).Thepresenceof>1%RPMI1640culturemediumleadstoasignalreductionduetothepresenceofexcessbiotinandironinthismedium.MEM,whichlacksthesecomponents,doesnotaffectAlphascreen®assays.
