Overview:
GSK3αisamultifunctionalproteinserinekinase,homologoustoDrosophila"shaggy"(zeste-white3)andimplicatedinthecontrolofseveralregulatoryproteinsincludingglycogensynthaseandtranscriptionfactors(e.g.,JUN)(1).GSK3αalsoplaysaroleintheWNTandPI3Ksignalingpathways.Alzheimerdiseaseisassociatedwithincreasedproductionandaggregationofamyloid-beta-40and-42peptidesintoplaques.GSK3αisrequiredformaximalproductionofthebeta-amyloid-40and-42peptidesgeneratedfromtheamyloidprecursorproteinbypresenilin-dependentgamma-secretasecleavage.Invitro,lithium,aGSK3αinhibitor,blockstheproductionofthebeta-amyloidpeptidesbyinterferingwiththegamma-secretasestep(2).
