CLIP-Surface™488isaphotostablegreenfluorescentsubstratethatcanbeusedtolabelCLIP-tag™fusionproteinsonthesurfaceoflivingcellsorinvitro.Thiscellimpermeablesubstrate(BC-488)isbasedonATTO-TECdyeATTO488andissuitableforstandardfluoresceinfiltersets.Ithasanexcitationmaximumat506nmandemissionmaximumat526nm.Thispackageincludes50nmolofCLIP-Surface488substrate,sufficienttomake10mlofa5µMCLIP-tag™fusionproteinlabelingsolution.TheCLIP-tag™proteinlabelingsystemenablesthespecific,covalentattachmentofvirtuallyanymoleculetoaproteinofinterest.CLIP-tagisaproteintagbasedonhumanO6-alkylguanine-DNA-alkyltransferase(hAGT).CLIP-tagsubstratesarederivativesofbenzylcytosine(BC).Inthelabelingreaction,thesubstitutedbenzylgroupofthesubstrateiscovalentlyattachedtothereactivecysteineofCLIP-tagformingastablethioetherbond.AlthoughCLIP-tagisbasedonthesameproteinasSNAP-tag,thebenzylcytosinesubstratesformaseparateclassofsubstrates,differentfromthebenzylcytosinesubstratesformaseparateclassofsubstrates,differentfromthebenzylguaninesubstratesrecognizedbySNAP-tag.CLIP-tagandSNAP-tag®canbeusedfororthogonalandcomplementarylabelingoftwoproteinssimultaneouslyinthesamecells.
