ThisassayisusedforthemeasurementofAβ(1-42)variantscleavedattheNterminalside.TheplaqueswhichappearinthebrainsofAlzheimer"sdiseasepatientsaremostlyconstitutedbytheAmyloid-βprotein(Aβ).Aβisapeptidewhichconsistsof40or42(43)aminoacids,andreportsshowthatthisiscleavedfromβ-andγ-secretasefromtheamyloidprecursorprotein(APP).ReportshaveshownmanyvariantsofAβexistandareclarifiedintotheculturesupernatantfromtheAPPCDNAtransfectedmouseneuroblastomacell.FurThermore,in1995,adominantanddifferentialdepositionofdistinctamyloid-βpeptidespecies,Aβ(N3pE),insenileplaqueswasfoundbySaidoetal.Thismodifiedmolecule,startingatthe3rdaminoterminalresidue,glutamate,wasdiscoveredtoconverttopyroglutamatethroughintramoleculardehydration.
