Thedetectionofproteinsinimmunoblottingapplicationsistypicallyachievedbyusinganantibodythatisconjugatedtoanenzyme,suchashorseradishperoxidase(HRP)oralkalinephosphatase(AP).Theprimaryantibodywhichdirectlybindstotheproteintargetmaybelabeledwiththeenzyme,oralabeledsecondaryantibodymaybeusedtobindtoanunlabeledprimaryantibody.Thecovalentlyattachedenzymeiscapableofdegradingasubstrate,whichmaybechromogenicorchemiluminescent.Chromogenicsubstratesaredegradedintocoloredprecipitatesandareeasytouse.Nospecialequipmentisrequiredfordetection.Chemiluminescentreactionsproducelightandaremoresensitivethanchromogenicreactions,buttheimagemustberecordedonx-rayfilmorwithageldocumentationsystem.Forbothtypesofsubstrates,thesignalgeneratedbytheenzymecatalyzedreactionisdetectedonthemembraneonlyatthesiteofthetargetprotein,iftheimmunoblottingconditionsareoptimized.
