WeighoutthedesiredamountofagaroseandplaceinanErlenmeyerflaskwithameasuredamountofelectrophoresisbuffer,e.g.foran0.8%gel,add0.8gmofagaroseand100mlofTBEBuffer(1X),toa200mlflask.Thelargerflaskinsuresagainsttheagaroseboilingover.Dissolvetheagaroseinaboilingwaterbathorinarevolving-platemicrowaveoven.Allthegrainsofagaroseshouldbedissolvedandthesolutionclear.Coolthesolutionto60°C(70°Cforconcentrations2%orabove)andpourimmediately.Allowthegeltosetforone-halfhourbeforeusing.Makesuretousethesameelectrophoresisbufferinthegelasfortherunningbuffer.
