TheTC20automatedcellcounterusesmicroscopywithauto-focusthatanalyzesmultiplefocalplanestoidentifythebestplane.Withoutrequiringanyuserinput,thesophisticatedcellcountingalgorithmusestheimageacquiredfromthebestfocalplanetoidentifycellsandexcludedebris,therebycalculatingthetotalcellcount.Theauto-focusleadstohighlyreproduciblecellcountswithreduceduser-to-uservariabilitycomparedtoahemocytometerandcellcounterswithmanualfocus.Usingauto-focusinsteadofsubjectivemanualfocusingisespeciallyimportantwhenassessingcellviabilitybecauseanincorrectlyselectedfocalplanewillleadtoinaccurateresults.

Unlikecellcountersbasedonelectricalimpedance(Coulterprinciple),theTC20image-basedanalysisallowstheusertoviewthecellsinrealtime,providingvisualproofthattheTC20cellcounterhascorrectlyidentifiedcells.

TheTC20automatedcellcounterprovidesaccuracycomparabletoresultsobtainedwithahemocytometer.Itcancountcellswitha6–50μmcelldiameterandwithinabroadconcentrationrangeof5x10⁴–1x10⁷cells/ml,whicheliminatestheneedtodilutecells,thusreducingerrorsassociatedwithsampledilutionspriortocounting.

TheTC20cellcounteruses multifocalplaneanalysistoassesscellviability.Theconventionalmethodofanalyzingviabilityusingasinglefocalplanecanleadtoinaccurateconclusionsbecauselightscatteringandthealignmentofcellsatdifferentheightsinacountingchambercanchangetheappearanceofcells—livecellsmayappeartobedeadandviceversa.Todetermineifcellsareviable,theTC20counteranalyzeseachcellusingimagesacquiredfrommultiplefocalplanesduringthefocusingstep.

ThebroadconcentrationrangeoftheTC20cellcountereliminatestheneedtodilutecellspriortocounting,reducingtheerrorsassociatedwithsampledilutionsthatmaybenecessarywhencountingcellsbyothermethods.Thecountingalgorithmdiscriminatesandcountsindividualcellswithinclustersofuptofivecells,providingaccuratecountswithouttheneedtoextensivelydeclumpcellspriortoloading.
