Thephosphorylationofmyosinaffectsitsroleinsmoothmusclecontraction,plateletformationandpossIBLyotherprocesses.Phosphorylationbymyosinlightchainkinase(MLCK)increasesmyosinactivityanddephosphorylationbymyosinphosphatasedecreasesmyosinactivity.CPI,afactorthatbindstoandinhibitsmyosinphosphatase,isatargetofphosphorylationbyPKCandPKN.TheinhibitoryactivityofCPIisregulatedbyitsownphosphorylationstate;whenCPIisphosphorylated,itsinhibitoryactivityisincreased.TheactivationofsignaltransductioncascadessuchasGPCRpathwayscanleadtoactivationofPKC,phosphorylationofCPI,inhibitionofmyosinphosphatase,increasedmyosinphosphorylationandincreasedsmoothmusclecontractionorplateletrelease.Theactionofhistamineinvasoconstriction,forexample,maybemediatedbyactivationofPKCthroughthehistaminereceptor,resultinginphosphorylationofCPI-17,increasedinhibitionofmyosinphosphatase,andincreasedsmoothmusclecontraction.
