Amprenavir(Agenerase,VX-478),isanorally-available,sulfonamide-basedinhibitorofHIV-1protease,withrelativelyweakpotencytowardsHIV-2,atIC50valuesof0.6and19nM,respectively.[1]SimilartootherHIV-1proteaseinhibitors,AmprenavirbindstotheactivesiteofHIV-1andinhibitstheprocessingofthegagandgag-polpolyproteinprecursors.ThedrugconcentrationofAmprenavirrequiredtoreduceviralproductionby50%(IC50)againstHIV-1strainIIIBwas0.08uMinacutelyinfectedcellsand0.41uMinchronicallyinfectedcells.UsingadifferentcelllinederivedfromahumanT-celllymphoma,theIC90ofAmprenaviragainstHIV-1strainIIIBwas0.079uM.[1]
Humandose-escalationstudiesshowedthatAmprenavirwaswelltoleratedatsingledosesupto1200mgwithminimaleffectonabsorptionbyfoodadmiNISTration,unlikewithotherproteaseinhibitorssuchasindinavir,nelfinavir,andsaquinavir.[2]
