Matchmaker Gold Yeast One-Hybrid Library Screening System은 미지의 단백질과 DNA 간의 상호작용을 확인하고 그 기능을 파악할 수 있는 간단하고 효율적인 방법을 제공한다 (Figure 1). 모든 Matchmaker Gold System은 정확하고 사용하기 편리한 reporter로 Aureobasidin A 내성 (AbAr)을 사용한다. 이 새로운 reporter는 항생제인 Aureobasidin A (AbA)로 인해 AbA 내성유전자를 발현하지 못하는 yeast를 효과적으로 제거하기 때문에 매우 낮은 screening background를 가능하게 하였다.
□ Matchmaker Gold One-Hybrid System
Matchmaker Gold Yeast One-Hybrid system에서는 목적 DNA (bait) 서열을 몇 번 반복하여 pAbAi reporter vector에 cloning 한 후, bait-specific reporter를 가진 yeast strain을 만들기 위하여 pAbAi vector를 homologous recombination으로 Y1HGold yeast genome으로 삽입시켜 library screening을 위한 숙주로 사용한다.

□ One-step Library 제작과 Screening
Yeast GAL4-transcription activation domain (GAL4 AD prey protein)과 융합되어 발현되는 DNA-binding protein을 포함하는 cDNA library는 SMART 기술과 homologous recombination을 이용하여 Y1HGold [Bait-AbAi] reporter strain에서 직접 제작할 수 있다. (Figure 2). Prey 단백질이 bait sequence와 결합하면 GAL4 AD는 AbA 저항성 유전자의 발현을 활성화시켜 AbA를 포함하는 배지에서 yeast가 생존하게 된다. 따라서 library screening 과정에서 살아남은 yeast clone으로부터 prey 단백질을 발현하는 plasmid를 쉽게 획득할 수 있고 이를 이용하여 다음 과정의 실험을 진행할 수 있게 된다.
□ Colony PCR Kit & Media Set
Colony PCR은 bait strain을 확인하고 screening을 통해 확인된 positive clone을 분류하기 위한 가장 빠르고 편리한 방법이다. Matchmaker Insert Check PCR Mix 1은 pBait-AbAi 재조합체가 yeast genome에 삽입되었는지 확인하기 위하여 사용하며, Matchmaker Insert Check PCR Mix 2는 onehybrid 또는 two-hybrid screening 과정에서 얻은 positive clone을 빠르게 분석하기 위하여 사용한다. Yeast Media Sets 1과 1 Plus는 Matchmaker Gold One-Hybrid 실험에 필요한 배지를 pouch 형태로 구성한 제품이다.
