RNAiMonitor는 RNAi 목적서열target sequences의 넉다운knockdown을 cell-lysis 과정 없이, 효과적이고 간편하게 모니터 할 수 있도록 설계된 제품이다.본 제품은 pRNAiMonitor-MetLuc2 Vector와 Ready-To-Glow Secreted Luciferase Reporter Assay제품으로 구성되어 있다.pRNAiMonitor-MetLuc2 vector는 MetLuc(분비되는 luciferase reporter)을 발현시켜, 세포 주변 배지를 이용해 간편하게 검출 가능하다. 목적서열(shRNA 또는 siRNA의 타겟을 코딩하는 유전자 서열)을 MetLuc유전자의 뒤쪽에 클로닝 한 후, MetLuc과 목적서열을 동시에 코딩하는 bicistronic mRNA 전사산물을 만든다. 목적유전자의 넉다운은 mRNA 전사산물의 분해(또는 translation 저해)를 야기하고, 리포터단백질의 발현을 감소시킨다. Ready-To-Glow Secreted Luciferase Reporter Assay kit을 이용하면 쉽게 리포터분자를 검출 할 수 있다.
□ Ready-To-Glow Secreted Luciferase Reporter Assay
Ready-To-Glow Secreted Luciferase Reporter Assay는 분비형 luciferase 유전자 Metridia longa 를 기반으로 만들어졌다. MetLuc는 N-terminal secretory signal peptide인 17개 아미노산잔기를 갖고 있는 24 kDa 리포터단백질이다(1). 이것은 luciferase 검출법의 민감도와 live-cell assay의 장점이 결합된 방법이다. 세포 lysis없이 트랜스펙션된 세포의 상층액(배지)로 분비된 리포터효소의 활성을 검출하여 RNAi의 레벨을 측정할 수 있는 원스텝 방법이다. 이 시그널signal이 안정적이어서 복수의 시료를 동시에 실험 할 수 있으며 기질substrate 첨가 후 시그널이 감소함에도 불구하고, 전반적인 유도발현 배율은 기질 첨가 후 30분 동안 거의 비슷한 수준으로 유지된다(2).
RNAiMonitor measures knockdown of RNAi target sequences efficiently and easily. A pMetLucCAV1 reporter plasmid (3 μg) containing a CAV1 target sequence, and the indicated amount of an anti-CAV1 shRNA expression vector, were cotransfected into HeLa cell cultures. A ProLabel expression vector was also included to assess transfection efficiency. At the end of the experiment, the culture medium was assayed for secreted luciferase activity, and the cells were assayed for ProLabel content. The level of luciferase knockdown correlated with the amount of cotransfected CAV1 shRNA construct.
The ProLabel Screening Kit allows you to quantitatively measure the level of any recombinant protein. The ProLabel tag and Enzyme Acceptor (EA) combine to form an active enzyme that cleaves the chemiluminescent substrate; the resulting signal can be detected with any standard luminometer.
