转染操作流程:1、细胞传代:
转染前24h内分离293T细胞至含10%胎牛血清的DMEM培养基中进行传代培养,接种密度如下:6孔板:0.5x106细胞10cm培养皿:4.0x106细胞15cm培养皿:9.0x106细胞
2、转染
转染前将所有试剂置于室温。
2.1质粒DNA的稀释
在无菌试管中,用无血清的DMEMW/O酚红培养基(浓度为10%)稀释质粒DNA(ug)。病毒包装体(psPAX2):病毒包膜(pMD2G)和重组质粒DNA的稀释比例分别为4:2:1。
6孔板:200ul+3ug的DNA10cm培养皿:1ml+7-8ug的DNA15cm培养皿:2ml+11-12ug的DNA2.2转染复合体形成
将PEI(1ug/uL)加入已稀释的总DNA中,PEI与DNA(ug)的混合比率为3:1。加入后立即涡旋混合。
6孔板:9ulPEI复合体(1ug/ul)=9ug10cm培养皿:21ulPEI复合体(1ug/ul)=21ug15cm培养皿:33ulPEI复合体(1ug/ul)=33ug将添加到细胞的DNA/PEI的混合物在室温下孵育15分钟。
4、收获转染细胞
转染后48小时内收获转染细胞/或病毒上清。试剂的配制:PEI(1ug/ul)polysciences(CAT#23966-2配制成储液:)1、将无内毒素的无菌水加热至80℃左右溶解PEI,冷却到室温。2、调整pH值至7.0,用0.22um的滤器过滤消毒,分装后储存在-20℃。工作液可保持在4℃
