Description
The mGluR1a Antibody is no longer available from ImmunoStar. To purchase this antibody, please refer to PhosphoSolutions. The antibody has significant biotin-avidin HRP staining at a 1/300 - 1/500 dilution in rat thalamus, hippocampus and cortex. Optimal dilution will vary depending upon fixation, labeling technique and/or detection system; therefore, a dilution series is recommended. The antibody is specific for the ~125k monomer and the ~250k mGluR1a dimer in Western blots of rat brain extracts. Immunolabeling for mGluR1a is completely abolished by preadsorption with synthetic rat mGluR1a (1116-1130) at 5 µg per mL of diluted antibody. Note: If needed, low levels of glutaraldehyde (0.1 - 0.3%) may be used in conjunction with paraformaldehyde.
Photo Description: IHC image of neurons staining for the mGluR1 alpha in the rat hippocampus (above) and western blot showing specificity of the antibody at 132 kDa (below). For IHC, the tissue was fixed with 4% formaldehyde in 0.1 M phosphate buffer, before being removed and prepared for vibratome sectioning. Floating sections were incubated at RT in 10% goat serum in PBS, before standard IHC procedure. Primary antibody was incubated at 1:500 for 48 hours, goat anti-rabbit secondary was subsequently added for 1 hour after washing with PBS. Light microscopy staining was achieved with standard biotin-streptavidin/HRP procedure and DAB chromogen.
