BIOLOGicalActivity:ML228isahypoxiainducIBLefactor(HIF)pathwayactivatorwithanEC50at~1.23µMinHREgenereporterassayand~1.4μMinHIF-1αnucleartranslocationassay.Itdoesnotinhibittheproteasome,butactivatesHIFstABIlizationandnucleartranslocation,andinducesexpressionofaHIFspecificdownstreamgene(VEGF).Ithasnoapparenttoxicitybelow30μMandappearedtobeanironchelator,independentlyofPHD.HIFsaretranscriptionfactorsresponsiblefortheactivationofgeneswhichencodeproteinsthatmediateadaptiveresponsestoreducedoxygenavailability.HIFpathwayisthemostsignificantpathwayforcellularresponsetohypoxia.
