Human placenta type alkaline phosphatase is a dimeric glycoprotein (N-linked glycans at asparagine
residues 144 and 271). The protein is bound to the cell-membrane via a GPI-anchor which is linked to
Aspartate 506.

Amino acid residues 23-511 of human placenta type alkaline phosphatase were cloned, the protein was
over-expressed and was puri?ed to homogeneity (?gure 1). The cDNA coding for the recombinant protein
does not contain the GPI-signal, resulting in a secreted, soluble form of placental alkaline phosphatase 1. The
calculated molecular weight of recombinant secretred human PLAP-1 is 54.5 kDa. The protein contains a Cterminal hexahistidine tag. Each vial contains 0.25 mg recombinant secreted human PLAP-1 with a speci?c
activity of 100 U/mg (determined with p-nitrophenylphosphate as a substrate in a glycine assay at
25 ?C).
Storage and stability
Recombinant secretred human PLAP-1 is sterile
and should be stored at 4 ?C (stable for at least
6 months). The bu°er contains 5 mM tris 2 mM
MgCl2 0.05 mM ZnCl2 1% mannitol pH 8.0
without preservative. After opening it should
be stored sterile in appropriate small aliquots
at 4 ?C.
