TheCultreCoat®VascularPermeABIlityAssayemploysasimplifiedBoydenchamberdesignwithaonemicronpolyethyleneterephthalate(PET)membranecoatedwithcollagenI.Portswithinthemigrationchamber(top)allowaccesstotheassaychamber(bottom)withoutdismantlingthedevice.Thisdesigniseasiertouse,preventscontamination,andisadaptableforrobotichighthroughputsystems.Theassaychambermaybedirectlyanalyzedinaplatereader,eliminatingtransferstepsthatintroduceadditionalvariabilitytotheassay.
Theconfluentcellmonolayerformsabarrierthroughcell-celladhesionmechanismssuchasjunctions(adherens,tight,andgap)anddesmosomes,andthisbarrierrestrictsFITC-Dextrantotheinserts.Disruptionofthesecell-celladhesionmechanismscreatesgapsintheintercellularspacethatallowdiffusionofFITC-Dextranintothebottomchamber,sotheamountofFITC-Dextranthatdiffusesisrelatedtotheamountofintercellularspace,whichisrepresentativeofvascularpermeability.
