Thetriplehelixisthehallmarkproteinstructureofcollagen.Duringtissueremodeling,thetriplehelicalcollagenmoleculesaredegradedbyspecificproteases(e.g.,MMPorcathepsinK)andbecomeunfoldedatbodytemperature.TheCollagenHybridizingPeptide(CHP)isasyntheticpeptidethatcanspecificallybindtosuchdenaturedcollagenstrandsthroughhydrogenbonding,bothinhistology,invivo,andinvitro(3Dcellculture).BysharingthestructuralmotifandtheGly-X-Yrepeatingsequenceofnaturalcollagen,CHPhasastrongcapABIlitytohybridizewithdenaturedcollagenstrands,inafashionthatissimilartoaDNAfragmentannealingtoitscomplimentaryDNAstrandduringPCR.CHPisanextremelyspecificprobeforunfoldedcollagenmolecules:ithasnegligIBLeaffinitytointactcollagenmoleculesduetothelackofbindingsites;itisalsoinerttowardsnon-specificbindingbecauseofitsneutralandhydrophilicnature.
