Apoptosis is known as programmed cell death implicated in biological processes ranging from embryogenesis to ageing and from normal tissue homoeostasis to many human diseases. A number of signaling pathways that lead to characteristic cell changes are involved in mediating the apoptosis responses. One critical change in signaling pathways is the activation of transcription factors (TFs), and TF-based reporters are often used to monitor the activation of signaling pathways. Principle:
Either the TF promotor (pLuc5U-Oxidative Stress Luciferase Reporter Vector Set) or the TF binding site (All other sets) is upstream of TATA promoter to induce the expression of luciferase gene. It results in the induction of luciferase enzyme activity. Through comparison of luciferase activity in two samples, either TF expression (pLuc5U-Oxidative Stress Luciferase Reporter Vector Set) or the up- or down-regulation of TFs can be quantitatively measured.
