Description:

Oxidative stress is caused by an imbalance in the generation and removal of reactive oxygen species (ROS).  ROS elicits a wide spectrum of responses, such as cell proliferation, growth arrest, or apoptosis. ROS comes mostly from the endogenous metabolic by-products and also from exogenous sources such as exposure to cigarette smoke, environmental pollutants, and bacterial, fungal or viral infections. Oxidative damage has been implicated in the cause of many diseases, such as cancer, Alzheimers disease, Parkinsons disease, atherosclerosis, and heart failure. A number of signaling pathways are involved in coordinating the response to elevations in ROS. One critical change in signaling pathways is the activation of transcription factors (TFs), and TF-based reporters are often used to monitor the activation of signaling pathways. 

Principle:

Either the TF promotor (pLuc5U-Oxidative Stress Luciferase Reporter Vector Set) or the TF binding site (All other sets) is upstream of TATA promoter to induce the expression of luciferase gene. It results in the induction of luciferase enzyme activity. Through comparison of luciferase activity in two samples, either TF expression (pLuc5U-Oxidative Stress Luciferase Reporter Vector Set) or the up- or down-regulation of TFs can be quantitatively measured.
