Antimicrobialproteins(AMP)areafundamentalelementoftheprimaryresponseagainstpathogens.AMPsaresmallendogenouscationicmoleculesexpressedbyphagocyticandepithelialcells.TheantimicrobialactivityofAMPsisdirectedtowardsabroadspectrumofpathogens,likeGram-positive&negativebacteria,viruses,yeastandfungi.AMPsaidininnateandadaptiveimmunityviadirectinactivationandbyimmunomodulatoryactivitylikeleukocytemigration.RNase7isanproteinsecretedbyavarietyofepithelialtissuesandisamemberoftheRNaseAsuperfamily.Thisfamilysharessequenceandstructuralsimilaritiessuchasconservedcysteineresiduesaswellasconservedhistidinesandalysineintheactivecentercatalysingtheribonucleaseactivity.RNase7isa14.5KDaproteinwithdistinctribonucleaseactivity.However,theantimicrobialactivityisindependentformtheribonucleaseactivitywhichmightbeassociatedwithantiviralactivity.Onapermolarbasis,RNase7isoneofthemostpotentAMPs.RNase7contributestothesterilityinseveralsystems.Itisdescribedtobeimportantinsterilityofthekidneyandurinarytractaswellasitscontributiontotheskinbarrierprotection.Intheurinarysystemitisconstitutivelyexpressedbytheintercalatedcellsintherenalcollectingtubulesandispresentintheurineatsuchlevelstokillbacteriaatbaseline.Intheskinitcontributestocontrolthegrowthofmicroorganismsontheskinsurface,andtheexpressionlevelscanbefurtherinducedundercontrolofproinflammatorycytokines.ThebactericidalactivityofRNase7hasbeenassociatedwithitsABIlitytobindandpermeatethebacterialcellmembrane.Thisrequiresclusteringoflysineresidues.RNase7isabletobindLPSandpeptidoglycans.Inocularsurface,signaltransductionassociatedwithRNase7expressionismediatedviaMAPKsbutnotNF-κBsignallingpathways.IL1βleadstoanincreasedexpressionofRNase7.

Principle | ThehumanRNase7ELISAisaready-to-usesolid-phaseenzyme-linkedimmunosorbentassaybasedonthesandwichprinciplewithaworkingtimeof3½hours.Theefficientformatofaplatewithtwelvedisposable8-wellstripsallowsfreechoiceofbatchsizefortheassay.SamplesandstandardsareincubatedinmicrotiterwellscoatedwithantibodiesrecognizinghumanRNase7.BiotinylatedtracerantibodywillbindtothecapturedhumanRNase7.Streptavidin-peroxidaseconjugatewillbindtothebiotinylatedtracerantibody.Streptavidin-peroxidaseconjugatewillreactwiththesubstrate,tetramethylbenzidine(TMB).Theenzymereactionisstoppedbytheadditionofoxalicacid.Theabsorbanceat450nmismeasuredwithaspectrophotometer.Astandardcurveisobtainedbyplottingtheabsorbance(linear)versusthecorrespondingconcentrationsofthehumanRNase7standards(log).ThehumanRNase7concentrationofsamples,whicharerunconcurrentlywiththestandards,canbedeterminedfromthestandardcurve. |
