DESCRIPTION:

Mono/di-nucleosomespurifiedfromchickenbloodcells.Thesearecorenucleosomescomprisinghistoneoctamers(twocopieseachofhistonesH3,H4,H2A,H2B),eachwrappedwith~146bpofDNAwith~50additionalbpofinternucleosomalDNA.Usefulfortheassayofvarioushistonemethyltransferases(e.g.MLL1Complex,MLL2Complex,MLL4Complex,Dot1LandNSD2)bymethodsemployingrADIolabelingwith[3H]-S-adenosylmethionine(e.g.gelelectrophoresis/autoradiographyorfilterplate/scintillationcounting).Mosthistonemethyltransferasesthatrequirenucleosomesforassayhaveroughlyequalorsomewhatgreateractivitywithchicken,relativetoHeLa,nucleosomes.

NOTE:Eachvialcontains50µgnucleosomalDNA,determinedbyA260nm.At~200bp/nucleosome,thetotalweight,DNA+protein,isthus~91µg.

A50ugvialofRBCnucleosomescontainsover80%moreproductthanthosefromvendorsthatsellbytotalweight.
