Superoxidedismutase(SOD)isanantioxidantenzymeinvolvedinthedefensesystemagainstreactiveoxygenspecies(ROS).SODcatalyzesthedismutationreactionofsuperoxiderADIcalanion(O2-)tohydrogenperoxide,whichisthencatalyzedtoinnocuousO2andH2Obyglutathioneperoxidaseandcatalase.SeveralclassesofSODhavebeenidentified.Theseincludeintracellularcopper,zincSOD(Cu,Zn-SOD/SOD-1),mitochondrialmanganeseSOD(Mn-SOD/SOD-2)andextracellularCu,Zn-SOD(EC-SOD/SOD-3)(1).SOD-1isfoundinalleukaryoticspeciesasahomodimeric32-kDaenzymecontainingoneeachofCuandZnionpersubunit(2).Themanganesecontaining80-kDatetramericenzymeSOD2,islocatedinthemitochondrialmatrixincloseproximitytoaprimaryendogenoussourceofsuperoxide,themitochondrialrespiratorychain(3).SOD-3isaheparin-bindingmultimerofdisulfide-linkeddimers,primarilyexpressedinhumanlungs,vesselwallsandairways(4).SOD-4isacopperchaperoneforsuperoxidedismutase(CCS),whichspecificallydeliversCutocopper/zincsuperoxidedismutase.CCSmayactivatecopper/zincsuperoxidedismutasethroughdirectinsertionoftheCucofactor.
