CREB(cAMPresponseelement-binding)proteinsaretranscriptionfactorswhichbindtocertainsequencescalledcAMPresponseelements(CRE)inDNA.Transcriptionfactorsplayakeyroleincontrollingcellproliferation,cellcycleprogression,fuelmetabolismandsoonthroughchangesintargetgenetranscription.CREBhasmanyfunctionsinmanydifferentorgansalthoughmostofitsfunctionshavebeenstudiedinrelationtothebrain.CREBproteinsinneuronsarethoughttobeinvolvedintheformationoflong-termmemories.CRE-BP1isoneofthemostabundantCRE-bindingproteins.ThelevelofitsmRNAisfairlyabundantinsomeregionsofthebrainsuchashippocampusandisinducedintheregeneratingliver,suggestingthatCRE-BP1isimportantforbothsignaltransductioninthebrainandcellularproliferation.Inthemajorityofpatientswithacutemyeloidleukemia(AML),CREBwasfoundtobeoverexpressedinthebonemarrowandthisoverexpressionwasassociatedwithpoorprognosis.CREB5(CRE-Bpa)consistsof508aminoacidsandhasamolecularweightof56,840.CREB5ishighlyhomologouswithCRE-BP1infourregions:twoofthemaretheregionscontainingtheputativemetalfingerortheDNA-bindingdomainconsistingofthebasicaminoacidclusterandtheleucinezipper.BindstothecAMPresponseelementandactivatestranscription.
