BACKGROUND

Thymidylate synthase (TS) is an intracellular enzyme critical for de novo synthesis of DNA. In cancer, expression of TS is often elevated and becomes further elevated as a result of treatment with the most commonly used chemotherapeutic, 5-fluorouracil (5-FU). Resistance or lack of response to 5-FU is attributed to the elevation of TS activity. Overexpression of TS has been demonstrated in many types of tumors including breast, colon, gastric and melanoma and is usually predictive of shortened survival time.

Antigen: Recombinant human thymidylate synthase produced in

E. coli.

SPECIFICITY

These antibodies specifically recognize human TS (36 kDa). They do not cross- react with rat TS.

APPLICATIONS

Immunohistochemistry: use at 5-10 ug/ml on frozen sections (reactivity on paraffin-embedded sections has not been determined).

Immunoblotting: use at 5-10 ug/ml. ELISA: use at 2-5 ug/ml (optimized for TS on solid phase at 5 ug/ml).

Immunoprecipitation: use #34002 only at 5-10 ug/ml.

DILUTION INSTRUCTIONS

Dilute in PBS or medium which is identical to that used in the assay system.

STORAGE AND STABILITY

This antibody is stable for at least one

(1) year at -20oC. Avoid multiple freeze- thaw cycles.

For in vitro investigational use only. Not for use in therapeutic or diagnostic procedures.
