BACKGROUND

Cellularresponsesinducedbythepro-inflammatorycytokineIL-1requireIL-1receptorcomplex(IL-1R1andIL-1RacP).Recently,MyD88wasidentifiedasanadaptermoleculeintheIL-1signalingpathway.MyD88associateswithandrecruitsIRAKtotheIL-1receptor.DominantnegativemutantsofMyD88attenuateIL-1R-mediatedNF-κBactivation.MyD88alsofunctionsasaregulatormoleculeforIL-18receptorandhumanTollreceptor,membersoftheToll/IL-1Rfamilyofreceptors.TargeteddisruptionoftheMyD88generesultsinlossofcellularresponsestoIL-1andIL-18,andMyD88-deficientmicelackresponsestoLPSwhichrequireToll-likereceptors2and4(TLR2andTLR4)asthesignalingreceptors.MyD88isageneraladapterproteinfortheToll/IL-1RfamilyofreceptorsandplaysanimportantroleintheinflammatoryresponsesinducedbycytokinesIL-1,IL-18,andLPS.MyD88isexpressedinavarietyoftissues.
