BACKGROUND

Hsp90isahighlyconservedandessentialstressproteinthatisexpressedinalleukaryoticcells.Itparticipatesinthefolding,assembly,maturation,andstABIlizationofspecificproteinsasanintegralcomponentofchaperonecomplexes.Hsp90isrelativelyabundantinunstressedcellsofmostprokaryoticandeukaryoticsystemsandcanbeinducedbyheatshockinsomesystems.Itexistsinadimericformandhasbeenobservedtobindtoseveralothercellularproteinssuchasretroviruskinases,steroidreceptors,hemeregulatedproteinkinase,actin,andtubulin.WhenboundtoATP,Hsp90interactswithco-chaperonescdc37,p23,andvariousimmunophilin-likeproteins,formingcomplexesthatstabilizeandprotecttargetproteinsfromproteasomaldegradation.
