Forty-twoantigenicdomainswereidentifiedacrossthehepatitisAvirus(HAV)polyproteinbyusingasetof237overlapping20-mersyntheticpeptidesspanningtheentireHAVpolyprotein.Nineteenantigenicdomainswerefoundwithinthestructuralproteins,and22werefoundwithinthenonstructuralproteins,with1domainspanningthejunctionofVP1andP2Aproteins.Fiveofthesedomainswereconsideredimmunodominant,asjudgedbyboththebreadthandthestrengthoftheirimmunoreactivity.OnedomainislocatedwithintheVP2proteinatposition57-90aa.Aseconddomain,locatedatposition767-842aa,containstheC-terminalpartoftheVP1proteinandtheentireP2Aprotein.Athirddomain,locatedatposition1403-1456a.a,comprisestheC-terminalpartoftheP2CproteinandtheN-terminalhalfoftheP3Aprotein.Thefourthdomain,locatedatposition1500-1519a.a,includesalmosttheentireP3Bandthelastdomain,locatedatposition1719-1764aa,containstheC-terminalregionoftheP3CproteinandtheN-terminalregionoftheP3Dprotein.FourofthefivemostimmunoreactivedomainsarederivedfromsmallHAVproteinsand/orencompassproteincleavagesitesseparatingdifferentHAVproteins.
