NeptuneBlockcontainsaproprietarynon-mammalianformulationbasedonfishproteins,plusmolecularstABIlizersthatprovideahighdegreeofblockingefficiency.NeptuneBlockwasdesignedforantigen-downELISAformats,andforsandwichassayswithhighbackgroundproblems.
Thisformulationisaheterogeneousmixtureofsmallmoleculescapableofblockingboththenonspecificbindingsitesoftheadsorbedprotein,aswellastheunoccupiedregionsofthepolystyreneplatethatarenotstericallyaccessIBLetolargertrADItionalmammalianblockingreagents,therebyreducingbackgroundnoise.Thesmallsizeoftheseuniqueblockersresultsinminimalsterichindrancetokeyepitoperegionsoftargetcoatedproteins,thusenhancingspecificsignalgeneration.
AsNeptuneBlockcontainsanon-mammalianproteinblockingagent,itisantigenicityforeigntomostmammalianimmunesystems.Inantigen-downELISAformats,thisreducesthepossibilityoffalse-positivesgeneratedfromendogenousantibodiesinthesamplereactingwithplateblockingproteins.NeptuneBlockisparticularlyusefulwhenworkingwithhuman,swine,andbovineserumsamples,asthereislittleinteractionwiththeblockingmoleculesresultinginlowerbackgrounds.
