mCherryisderivedfromproteinsoriginallyisolatedfromCnidarians(jellyfish,seaanemonesandcorals),andisusedasafluorescenttracerintransfectionandtransgenicexperiments.TheprototypeforthesefluorescentproteinsisGreenFluorescentProtein(GFP),whichisa~27kDaproteinisolatedoriginallyfromthejellyfishAequoriavictoria.ThemCherryproteinisderivedfromDsRed,aredfluorescentproteinrelatedtoGFPisolatedfromso-calleddisccoralsofthegenusDiscosoma.DsRedissimilarinsizeandpropertiestoGFP,but,obviously,producesaredratherthanagreenFluorochrome.

Severalfurthercyclesofmutation,directedmodificationandevolutionaryselectionproducedmCherry,whichhasanexcitationmaximumat587nmandandemissionmaximumat610nm(4).WeexpressedthemCherryproteinsequenceshowninreference4inbacteria,purifiedoutthemCherryandraisedthisrabbitpolyclonalantibody.ThiswasaffinitypurifiedandwasfoundtostainabandoftheexpectedsizeinHEK293cellstransfectedwiththepFin-EF1-mCherryvectordesignedtoexpressmCherry.Asshownbelow,theantibodydoesnotstainanyproteinbandinuntransfectedHEK293cells.
