Thespecificityoftheantiserumwasexaminedbysolublepre-adsorptionwiththepeptidesinquestionatafinalconcentrationof10-6MCCK-8immunolabelingwascompletelyabolishedbypre-adsorptionwithCCK-8,gastrin17andgastrin34.Pre-adsorptionwiththefollowingpeptidesresultedinnoreductionofimmunostaining:α-CGRP,Ÿ-CGRP,neurotensin,somatostatin,substanceP,leucineenkephalin,methionineenkephalin,VIP,neuropeptideY,gastricinhibitorypolypeptide,bombesin,glucagon,peptideYY,andFMRFamide.
ProductDescription:LowmagnificationIHCimageofneuronsstainingforCCK-8inthedorsalhornoftheratspinalcord(above),andintherathippocampus(below).Thetissuewasfixedwith4%formaldehydeinphosphatebuffer,beforebeingremovedandpreparedforvibratomesectioning(hippocampus)andfrozensectioning(spinalcord).SectionswereincubatedatRTin10%goatseruminPBS,beforestandardIHCprocedure.Primaryantibodywasincubatedat1:1000for48hours,goatanti-rabbitsecondarywassubsequentlyaddedfor1hourafterwashingwithPBS.Lightmicroscopystainingwasachievedwithstandardbiotin-streptavidin/HRPprocedureandDABchromogen.
