DescriptionmCherryisanengineeredderivativeofoneofafamilyofproteinsoriginallyisolatedfromCnidarians(jellyfish,seaanemonesandcorals).ThemCherryproteinwasderivedfromDsRed,aredfluorescentproteinfromso-calleddisccoralsofthegenusDiscosoma.DsRedisa223aminoacid~28kDaproteinsimilarinsizeandpropertiestoGFP,but,obviously,producesaredratherthanagreenFluorochrome.TheoriginalDsRedwasengineeredextensivelyintheTsienlabtopreventitfromformingtetramersanddimersandtomodifyandimprovethespectralproperties(1-3).TheresultingmonomericproteinisusefulforapplicationssuchasFörsterResonanceEnergyTransfer(FRET,alsoknownasFluorescenceResonanceEnergyTransfer).Severalfurthercyclesofmutation,directedmodificationandevolutionaryselectionproducedmCherry,whichismonomericandhasanexcitationmaximumat587nmandandemissionmaximumat610nm(4).
