Amylo-Glo®RTD™“ReadytoDilute”Stainingreagentisdesignedtostainamyloidplaquesintissuesections.ThisnovelMarkerhasseveraladvantagesoverotherconventionalmarkerssuchasThioflavinSandCongoRedbecauseofitsuniquechemicalandspectralproperties.(L.Schmuedetal.(2012)J.NeuroscienceMethods209:120–126).  UsingAmylo-Glo®resultsinaverybrightblueUVexcitablestainunderphysiologicalconditionsthatwillnotbleedthroughwhenilluminatedwithotherfilters.Itsbrightnessmakesitidealforlowmagnificationquantificationstudies,whileitsuniqueexcitation/emissionprofileandmildstainingconditionsmakesitidealforcombinationformultipleimmunofluorescentlabelingstudies.Amylo-Glo®RTD™iscompatIBLewithfresh,frozen,andformalin-fixedimmunohistochemistryorcytochemistry,anditisparticularlygoodforconfocalandmultiplelabelingbecauseofitshighfluorescentintensityandhighresistancetophoto-bleaching.MoreoverbecauseAmylo-Glo®fluorescesintheUVchannel,doubleandtriplelabelingexperimentscanbeperformedveryeasily(seeprotocol).
