N-terminalFLAG-taggedhumanPDE7Ahasbeenstablyexpressedinahumanembryonickidney(HEK293)cellline.PDE7AexpressionwasconfirmedbyWesternblotting.TheregulationofintracellularlevelofcAMPbyPDE7AinPDE7Astably-expressedHEK293cellswascharacterizedbyacell-basedreporterassayusingpCRE-lucreportervector.pCRE-luccontainsaluciferasegenethatisunderthecontrolofthecAMPresponseelement(CRE).WhencellstransientlytransfectedwithpCRE-lucreporterwereactivatedbyforskolin,thelevelofcAMPwasupregulatedinparentalHEK293cells,inducingtheexpressionoftheluciferasereporter,whereashPDE7A-HEK293cellsshowedreductioninthelevelofcAMPthatresultedinloweredexpressionofluciferase.InhibitionofPDE7AactivitybyBRL50481,aPDE7Ainhibitor,restoredthecAMPlevel,resultinginhigherluciferaseactivity.
