EZH2(UniProt/Swiss–ProtentryQ15910)isahistone–lysinemethyltransferasewhichmethylatesLys–9andLys–27ofhistoneH3,leADIngtotranscriptionalrepression.Itisamemberofthepolycombgroup(PcG)familywhichformmultimericproteincomplexesandareinvolvedinmaintainingthetranscriptionalrepressivestateofgenesoversuccessivecellgenerations.TheEZH2activityisdependentontheassociationwithothercomponentsofthePRC2complex(EED,SUZ12/JJAZ1,RBBP4andRBBP7).EZH2mayplayaroleinthehematopoieticandcentralnervoussystems.Over–expressionofEZH2isobservedduringadvancedstagesofprostatecancerandbreastcancer.

ChIPresultsobtainedwiththeantibodydirectedagainstEZH2
ChIPassayswereperformedusingHeLacells,theantibodyagainstEZH2(Cat.#25223)andoptimizedPCRprimersetsforqPCR.ChIPwasperformedusingshearedchromatinfrom1millioncells.Atitrationoftheantibodyconsistingof0.5,1,2.5and5µgperChIPexperimentwasanalyzed.IgG(2µg/IP)wasusedasnegativeIPcontrol.QuantitativePCRwasperformedwithprimersforMYT1,usedasapositivecontroltarget,andforthepromoteroftheactiveGAPDHgene,usedasanegativecontrol.Figure1showstherecovery,expressedasa%ofinput(therelativeamountofimmunoprecipitatedDNAcomparedtoinputDNAafterqPCRanalysis).

WesternblotanalysisusingtheantibodydirectedagainstEZH2
NuclearextractsofHeLacells(40µg)wereanalyzedbyWesternblotusingtheantibodyagainstEZH2(Cat.#25223)diluted1:1,000inTBS-Tweencontaining5%skimmedmilk.Thepositionoftheproteinofinterest(expectedsize85kDa)isindicatedontheright;theMarker(inkDa)isshownontheleft.
