Background:
L-tryptophan(L-Trp)isanessentialaminoacidnecessaryforproteinsynthesisinmammaliancellsandtheL-Trptokynurenine(Kyn)pathwayisfirmlyestablishedasakeyregulatorofinnateandadaptiveimmunity.CatabolismofL-TrptoKynmaintainsanimmunosuppressivemicroenvironmentbystarvingimmunecellsofL-TrpandreleasingdegradationproductsofL-Trpthathaveimmunosuppressivefunctions.Tryptophan2,3dioxygenase(TDO),aratelimitingenzymesinthispathway,isupregulatedinmanytumors,providingcancercellswithanavenueforimmuneevasion.
Description:
TheTDOFluorogenicInhibitorScreeningAssayKitisdesignedtomeasureenzymeinhibitionoftryptophan2,3-dioxygenase(TDO).TheTDOFluorogenicInhibitorScreeningAssayKitissimpletouseanddetectsfluorescenceatlongwavelengths,whichminimizespotentialerrorsduetocompoundinterference.Intheassay,theinhibitorandenzymeareaddedtoasamplecontainingL-Trpsubstrate.Aftera1hourincubationatroomtemperature,thefluorescencesolutionisaddedandincubatedat37°Cforfourhours.ActivityismeasuredbyreADIngsamplefluorescenceat&lamBDa;=510nmfollowingexcitationofthereactionproductatλ=400nm.
