Description:
TheLSD1AssayKitisdesignedtomeasureLSD1activityofpurifiedLSD1enzymescontainingLSD1usingfluorimetry.TheassayisbasedontheanalysisofthehydrogenperoxideproducedduringthedemethylationreactionmediatedbyLSD1.ThekeytotheLSD1AssayKitisthemethylatedpeptidesubstrate.Usingthiskit,onlytwosimplestepsonamicrotiterplatearerequiredforLSD1reactions.First,asamplecontainingLSD1isincubatedinareactionmixturewiththemethylatedpeptidesubstrate.Second,peroxidaseandAmplexRedreagentareaddedandfluorescence(λex=530±13nm,λem=590±18nm)ismeasuredusingaplatereader.
