TheJMJD1AHomogeneousAssayKitisdesignedtomeasureactivityoftheJMJD1Aforscreeningandprofilingapplications.JMJD1A,alsoknownasKDM3A,isaJumonjiC(JmjC)domain-containinghistonelysinedemethylasethatexhibitsdemethylationactivitytowardmonoanddimethyl-lysine9onhistoneH3.ThekeytotheJMJD1AHomogeneousAssayKitisahighlyspecificantibodythatrecognizesdemethylatedsubstrate.Withthiskit,onlythreesimplestepsonamicrotiterplatearerequiredfordemethylaseactivitydetection.First,asamplecontainingJMJD1Aenzymeisincubatedwiththebiotinylatedsubstrate.Next,acceptorbeadsandprimaryantibodyareadded,thendonorbeads,followedbyreADIngtheAlpha-counts.

Contraindications:
GreenandbluedyesthatabsorblightintheAlphaScreensignalemissionrange(520-620nm),suchasTrypanBlue.Avoidtheuseofthepotentsingletoxygenquencherssuchassodiumazide(NaN3)ormetalions(Fe2+,Fe3+,Cu2+,Zn2+andNi2+).ThepresenceofculturemediumRPMI1640at>1%leadstosignalreductionduetothepresenceofexcessbiotinandironinthismedium.MEM,whichlacksthesecomponents,doesnotaffectAlphaScreenassays.
