Lysyl-endopeptidase(Lys-c)wasisolatedfromtheGram-negativesoilbacteriumAchromobacterlyticusbyMasakietal.TheproteinhydrolyzesamideandpeptideesterbondsatthecarboxylicsideoflysineandS-aminoethylcysteineresiduesmakingitanimportanttoolforenzymaticproteinsequencingandLys-Xcompoundsynthesis.Theenzymefunctionsoptimallybetween30–37oCandsuffersfromdegradationwhensubjectedtotemperaturesabove50oC.Lysyl-endopeptidaseretainscompleteactivityafterincubationin4Mureaorin0.1%SDSsolutionforupto6hoursat30oC.Conversely,theenzymeisinhibitedbyDFP,PMSF,TLCK.Lysyl-endopeptidaseisinvolvedduringtheproductionofrecombinantinsulinandinproteomicsresearch.ImmobilizedLysyl-Endopeptidaseiscovalentlycross-linkedtoanagarosematrixcircumventingtheneedforenzymeremovalaftercleavage.Theresincanbereused10-20timesafterregeneratingwhenproperstorageconditionsarefollowed.
