Overview:
PDK1(3-phosphoinositide-dependentproteinkinase)isactivatedbythepresenceofPtdIns(3,4,5)P3orPtdIns(3,4)P2(1).PDK1thenactivatesproteinkinaseB(PKB)which,inturn,inactivatesglycogensynthasekinase-3(GSK3).PhosphorylationofotherproteinsbyPKBandGSK3islikelytomediatemanyoftheintracellularactionsofinsulin.Thus,PDK1playsakeyroleinmediatingmanyoftheactionsofthesecondmessenger(s)PtdIns(3,4,5)P3and/orPtdIns(3,4)P2.ThehumanPDK1isa556-residuemonomericenzymecomprisingofacatalyticdomainthatismostsimilartothePKA,PKBandPKCsubfamilyofproteinkinases.PDK1activityisdependentonphosphorylationatSer241.OtherknownphosphorylationtargetsitesincludeSer25,Ser393,Ser396andSer410(3).
