Theepidermalgrowthfactorreceptor(EGFR)isatransmembranetyrosinekinase.KnownligandsofEGFRincludeEGF,TGFA/TGF-alpha,amphiregulin,epigen/EPGN,BTC/betacellulin,epiregulin/EREGandHBEGF/heparin-bindingEGF.Ligandbindinginducesreceptorhomo-andhetero-dimerizationandtyrosineautophosphorylation,whichtriggersdownstreamsignalingevents.TheconsequencesofEFGRsignalingincludecellproliferation,differentiation,motility,andcellsurvival(1).ActivationofEGFRtriggersmitogenicsignalingingastrointestinalmucosaandEGFRupregulatedincoloncancersandmostneoplasms(2).EGFRstimulationalsoactivatestheERK-signalingpathwayinnormalgastricepithelialandcoloncancercelllines.Incontrast,selectiveinhibitionofEGFRsignificantlyreducesERK2activation,c-fosmRNAexpressionandcellproliferation.MutationsinEGFRarealsoimplicatedinspecificformsoflungcancer.
