Beta-cateninsareubiquitouslyexpressedcytoplasmicproteins.TheyassociatewithE-cadherinandthusrepresentcentralcomponentsofthecadherincelladhesioncomplex(1).beta-catenininteractswithTCFandLEFtranscriptionfactorsandareessentialforneuraldevelopmentintheWingless/Wntsignalingpathway(2).Theadenomatouspolyposiscoli(APC)tumor-suppressorprotein,togetherwithAxinandGSK3beta,formaWnt-regulatedsignalingcomplexthatmediatesphosphorylationdependentdegradationofbeta-cateninbytheproteasome.Specifically,beta-cateninisregulatedbysequentialphosphorylationofSer29,Ser33,Ser37andThr41byglycogensynthasekinase3beta(GSK3beta)(Liuetal.2002).Thishyperphosphorylationpromotestheubiquitylationandtargeteddegradationofbeta-catenin.Mutationsincomponentsofthisphosphorylationregulatedprocessthatpreventbeta-cateninhyperphosphorylationbyGSK3betaareassociatedwithcancers(4).
