The amount of cells that migrated through to the lower chemotaxis chamber was measured by Resazurin (Catalog&amp;nbsp;# AR002). Chemotaxis elicited by Recombinant Human IL-8/CXCL8 (20&amp;nbsp;ng/mL) is neutralized (green line) by increasing concentrations of Mouse Anti-Human IL-8/CXCL8 Monoclonal Antibody (Catalog&amp;nbsp;# MAB208). IL-8/CXCL8 was detected in immersion fixed human peripheral blood mononuclear cells (PBMCs) using Mouse Anti-Human IL-8/CXCL8 Monoclonal Antibody (Catalog # MAB208) at 10&amp;nbsp;µg/mL for 3 hours at room temperature. Formalin fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 10 minutes at room temperature. Cells were then blocked with 1% Blocker BSA for 15 minutes at room temperature. Cells were probed with a mouse monoclonal antibody recognizing STAT3, at a dilution of 1:100 for at least 1 hour at room temperature. Cells were then washed with PBS and incubated with DyLight 488 goat-anti-mouse secondary antibody at a dilution of 1:400 for 30 minutes at room temperature. Nuclei (blue) were stained with Hoechst 33342 dye.

Western blot shows lysates of ME-180 human cervical epithelial carcinoma cell line, HT-2 mouse T cell line, and Rat-2 rat embryonic fibroblast cell line. PVDF membrane was probed with 0.5&amp;nbsp;µg/mL Goat Anti-Human/Mouse/Rat CSRP1 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF5739) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog&amp;nbsp;# HAF109). For additional reference, recombinant human CSRP1, CSRP2, CSRP3 (5&amp;nbsp;ng/lane) were included. A specific band for CSRP1 was detected at approximately 23 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.
