Background
GM‑CSFwasinitiallycharacterizedasafactorthatcansupporttheinvitrocolonyformationofgranulocyte‑macrophageProgenitors.Itisalsoagrowthfactorforerythroid,megakaryocyte,andeosinophilprogenitors.GM‑CSFisproducedbyanumberofdifferentcelltypes(includingTcells,Bcells,macrophages,mastcells,endothelialcells,fibroblasts,andADIpocytes)inresponsetocytokineorinflammatorystimuli.Onmaturehematopoieticcells,GM‑CSFisasurvivalfactorforandactivatestheeffectorfunctionsofgranulocytes,monocytes/macrophages,andeosinophils(1,2).GM‑CSFpromotesaTh1biasedimmuneresponse,angiogenesis,allergicinflammation,andthedevelopmentofautoimmunity(3‑5).Itshowsclinicaleffectivenessinamelioratingchemotherapy‑inducedneutropenia,andGM‑CSFtransfectedtumorcellsareutilizedascancervaccines(6,7).The22kDaglycosylatedGM‑CSF,similartoIL‑3andIL‑5,isacytokinewithacoreoffourbundledalpha‑helices(8‑12).MaturehumanGM‑CSFshares63%‑70%aminoacidsequenceidentitywithcanine,feline,porcine,andratGM‑CSFand54%withmouseGM‑CSF.GM‑CSFexertsitsBIOLOGicaleffectsthroughaheterodimericreceptorcomplexcomposedofGM‑CSFRalpha/CD116andthesignaltransducingcommonbetachain(CD131)whichisalsoacomponentofthehigh‑affinityreceptorsforIL‑3andIL‑5(13,14).Inaddition,GM‑CSFbindsanaturallyoccurringsolubleformofGM‑CSFRalpha(15).HumanGM‑CSFisactiveoncanineandfelinecellsbutnotonmurinecells(16‑18).
